When it comes to the fastest heroes in the DC Comics universe, it goes without saying that the Flash --- be it Jay Garrick, Wally West, or Barry Allen --- is among the fastest of the fast. He's held his own against similar speedsters, be they hero or villain, and has even managed to outrun Superman on more than one occasion. But that doesn't mean that Kryptonians are without their super-speed too; so what actually happened when Barry Allen went toe-to-toe with one of the other famous Kryptonians, Superman's cousin herself: Supergirl? The result is one of the shortest --- yet fastest --- fights between two super-powered characters in recent publishing history.
It all happens in the pages of Supergirl #16, during the "H'El On Earth" storyline. Here, Supergirl has been convinced by the Kryptonian villain, H'El, that Superman has been actively suppressing bringing Krypton and its culture back into existence, by trying to stop H'El from going back in time and stopping the planet's destruction. Viewing himself as the sole keeper of the future of Krypton, H'El has found himself an ally in the form of Supergirl, who has kept herself within the Fortress of Solitude. While the rest of the Justice League is off handling other matters in a different part of the Fortress, the Flash finds himself sneaking into the Fortress by phasing through its walls.
Within the Fortress he confronts Supergirl, convinced still that the Flash is an enemy along with Superman. H'El's influence on Kara is marked by her seemingly being unable to remember Barry Allen, his abilities, or his incredible speed. As the Flash tries desperately to reason with her, they tussle, and in the span of a single splash page, engage in a heated battle. Even as the two heroes fly at each other, Supergirl notes how it is the fastest battle in which she's ever been engaged. Since both are moving at such incredible speeds, and despite her immense power, she holds back a bit, so they're nearly equally matched, and although to the naked eye it would appear to be over in a second, they're both able to attempt a plethora of various tactics against each other.
Passing through the Fortresses many rooms, the pair find themselves in a menagerie, where together they both subdue several of the more aggressive alien creatures, seemingly to help one another...that is, until Supergirl and the Flash find themselves in one of the Fortress's many corridors, having fought their way through the animal menagerie, and come face-to-face with one more ferocious creature: Krypto, the Superdog. Tossed aside once more by now two Kryptonian threats, the Flash manages to find himself in the Fortress's armory, where --- as instructed by Superman himself --- he grabs a weapon designed specifically against Kryptonians. Flash is able to use the device only briefly before H'El arrives, breaking apart the device, and subduing the Flash.
Despite the speed at which this fight between Supergirl and Flash plays out, it is still quite brutal and consequential. Here, Supergirl is barely able to keep pace with the Scarlet Speedster, as her loss of memory of him means she can't recall just HOW fast he can be. Flash, for his part, uses his myriad abilities to keep Supergirl at bay for long enough to keep her confused (phasing the both of them through the Fortress's walls keeps her considerably confused), but ultimately he just can't handle more than one Kryptonian at a time. While he winds up on the ground at the end, and while they share at least one super-power, this super-speed brawl keeps both the Flash and Supergirl at odds...in the fastest, shortest fight imaginable.
